Aciclovir is a synthetic purine nucleotide analogue, which acts against human herpes viruses. The enzyme thymidine kinase (TK) encoded by HSV, VZV and EBV converts acyclovir into acyclovir monophosphate, a nucleotide analogue, which is further converted to diphosphate and triphosphate by cellular enzymes. Acyclovir triphosphate interferes with DNA polymerase and inhibits viral DNA replication with resultant chain termination following its incorporation into the growing viral DNA chain.
